Abstract San Basilio de Palenque is an Afro-descendant community near Cartagena, Colombia, founded in the sixteenth century. The recognition of the historical and cultural importance of Palenque has promoted several studies, namely concerning the African roots of its first inhabitants. To deepen the knowledge of the origin and diversity of the Palenque parental lineages, we analysed a sample of 81 individuals for the entire mtDNA Control Region as well as 92 individuals for 27 Y-STRs and 95 for 51 Y-SNPs. The results confirmed the strong isolation of the Palenque, with some degree of influx of Native American maternal lineages, and a European admixture exclusively mediated by men. Due to the high genetic drift observed, a pairwise F ST analysis with available data on African populations proved to be inadequate for determining population affinities. In contrast, when a phylogenetic approach was used, it was possible to infer the phylogeographic origin of some lineages in Palenque. Contradicting previous studies indicating a single African origin, our results evidence parental genetic contributions from widely different African regions.

Résumé San Basilio de Palenque est une communauté d'ascendance africaine près de Cartagena, en Colombie, fondée au XVIe siècle. La reconnaissance de l'importance historique et culturelle de Palenque a favorisé plusieurs études, notamment sur les racines africaines de ses premiers habitants. Pour approfondir la connaissance de l'origine et de la diversité des lignées parentales de Palenque, nous avons analysé un échantillon de 81 individus pour l'ensemble de la région de contrôle de l'ADNmt ainsi que 92 individus pour 27 Y-STR et 95 pour 51 Y-SNP. Les résultats confirment le fort isolement des Palenque, avec un certain afflux de lignées maternelles amérindiennes, et un mélange européen exclusivement médiatisé par les hommes. En raison de la forte dérive génétique observée, une analyse F ST par paires avec les données disponibles sur les populations africaines s'est avérée inadéquate pour déterminer les affinités entre les populations. El reconocimiento de la importancia histórica y cultural de Palenque ha impulsado varios estudios, a saber, sobre las raíces africanas de sus primeros habitantes. Para profundizar en el conocimiento del origen y la diversidad de los linajes parentales de Palenque, analizamos una muestra de 81 individuos para toda la Región de Control de ADNmt, así como 92 individuos para 27 Y-STR y 95 para 51 Y-SNP. Los resultados confirmaron el fuerte aislamiento del Palenque, con cierto grado de afluencia de linajes maternos nativos americanos, y una mezcla europea exclusivamente mediada por hombres. Debido a la alta deriva genética observada, un análisis F ST por pares con datos disponibles sobre poblaciones africanas demostró ser inadecuado para determinar las afinidades de la población.
