Glycaemic, Lipidaemic and Antioxidant Profiles of Alloxan-Induced Diabetic Wistar Rats Treated with Glibenclamide and Aqueous Extract of <i>Gongronema latifolium</i> (Benth)

The current study investigated the ameliorative effects of combined therapy of glibenclamide and G. latifolium (GL) on several biochemical parameters of alloxaized Wistar rats. Thirty adult male Wistar rats assigned into 5 groups of 6 rats each were used for the study. Groups 2-5 were intraperitoneally injected with 160 mg/kg of alloxan monohydrate and upon establishment of diabetes (Fasting Blood Glucose (FBG) ≥ 126 mg/dl) were treated with 10 ml/kg distilled water (DW), 2 mg/kg glibenclamide, 200 mg/kg GL and 2 mg/kg glibenclamide and 200 mg/kg GL respectively. Rats in group 1 were not made diabetic and served as normal control. All the treatments were realized through daily oral route using gastric tube, for 21 days. Results indicated that the treatment of diabetic rats with a combination of glibenclamide and GL significantly reduced the elevated glucose levels, cholesterol, triacylglycerol, low density lipoprotein and malondialdehyde levels, along with increases in the high density lipoprotein, glutathione values and catalase activities, when compared to diabetic untreated group. It was concluded that the combined therapy of glibenclamide and GL showed superior antihyperglycemic, hypolipidaemic and antioxidant effects compared to either of them used alone.

La présente étude a examiné les effets améliorateurs d'une thérapie combinée de glibenclamide et de G. latifolium (GL) sur plusieurs paramètres biochimiques de rats Wistar alloxaisés. Trente rats Wistar mâles adultes répartis en 5 groupes de 6 rats chacun ont été utilisés pour l'étude. Les groupes 2 à 5 ont été injectés par voie intrapéritonéale avec 160 mg/kg d'alloxane monohydraté et lors de l'établissement du diabète (glycémie à jeun (FBG) ≥ 126 mg/dl) ont été traités avec 10 ml/kg d'eau distillée (DW), 2 mg/kg de glibenclamide, 200 mg/kg de GL et 2 mg/kg de glibenclamide et 200 mg/kg de GL respectivement. Les rats du groupe 1 n'ont pas été rendus diabétiques et ont servi de contrôle normal. Tous les traitements ont été réalisés par voie orale quotidienne par sonde gastrique, pendant 21 jours. Les résultats ont indiqué que le traitement des rats diabétiques avec une combinaison de glibenclamide et de GL réduisait significativement les taux élevés de glucose, de cholestérol, de triacylglycérol, de lipoprotéines de faible densité et de malondialdéhyde, ainsi que les augmentations des lipoprotéines de haute densité, des valeurs de glutathion et des activités catalasiques, par rapport au groupe diabétique non traité. El presente estudio investigó los efectos de mejora de la terapia combinada de glibenclamida y G. latifolium (GL) en varios parámetros bioquímicos de ratas Wistar aloxaizadas. Para el estudio se utilizaron treinta ratas Wistar macho adultas asignadas a 5 grupos de 6 ratas cada uno. Los grupos 2-5 se inyectaron por vía intraperitoneal con 160 mg/kg de monohidrato de aloxano y, tras el establecimiento de la diabetes (glucosa en sangre en ayunas (FBG) ≥ 126 mg/dl), se trataron con 10 ml/kg de agua destilada (DW), 2 mg/kg de glibenclamida, 200 mg/kg de GL y 2 mg/kg de glibenclamida y 200 mg/kg de GL, respectivamente. Las ratas del grupo 1 no se convirtieron en diabéticas y sirvieron como control normal. Todos los tratamientos se realizaron por vía oral diaria mediante sonda gástrica, durante 21 días. Los resultados indicaron que el tratamiento de ratas diabéticas con una combinación de glibenclamida y GL redujo significativamente los niveles elevados de glucosa, colesterol, triacilglicerol, lipoproteína de baja densidad y malondialdehído, junto con aumentos en la lipoproteína de alta densidad, valores de glutatión y actividades de catalasa, en comparación con el grupo diabético no tratado.
