Halogen Dance on 2‐Iodobenzofuran and 2‐Iodobenzothiophene and Related Reactions

Abstract When 2‐iodobenzofuran was treated sequentially with lithium 2,2,6,6‐tetramethylpiperidide in tetrahydrofuran at −50 °C and an aldehyde, the 2‐substituted 3‐iodobenzofuran resulting from the halogen dance was the only isolated product. However, from 2‐iodobenzothiophene, these conditions led to mixtures in which the 2‐substituted 3‐iodobenzothiophene was always accompanied by the 2‐substituted benzothiophene. The use of 2‐bromobenzothiophene as a catalyst made it possible to significantly reduce this competitive dehalogenation. To confirm the halogen dance reaction, the products were unambiguously synthesized by using direct halogenations and deprotolithiation‐trapping sequences as key steps. Our efforts to access 2,7‐disubstituted and 7‐substituted derivatives of benzofuran and benzothiophene have also been reported.

Résumé Lorsque le 2‐ iodobenzofurane a été traité séquentiellement avec du 2,2,6,6‐tétraméthylpipéridide de lithium dans du tétrahydrofurane à −50 °C et un aldéhyde, le 3‐iodobenzofurane 2-substitué résultant de la danse halogène était le seul produit isolé. Cependant, à partir du 2‐ iodobenzothiophène, ces conditions ont conduit à des mélanges dans lesquels le 3‐ iodobenzothiophène2 ‐ substitué était toujours accompagné du benzothiophène 2‐substitué. L'utilisation du2-bromobenzothiophènecomme catalyseur a permis de réduire significativement cette déshalogénation compétitive. Resumen Cuando el 2 iodobenzofurano se trató secuencialmente con 2,2,6,6‐tetrametilpiperidida de litio en tetrahidrofurano a -50 °C y un aldehído, el 3 ‐ yodobenzofurano 2 ‐ sustituido resultante de la danza halógena fue el único producto aislado. Sin embargo, a partir de 2 iodobenzotiofeno, estas condiciones condujeron a mezclas en las que el 3 iodobenzotiofeno 2-sustituido siempre estuvo acompañado por el benzotiofeno 2-sustituido. El uso de2-bromobenzotiofeno como catalizador permitió reducir significativamente esta deshalogenación competitiva. Para confirmar la reacción de baile halógeno, los productos se sintetizaron sin ambigüedades mediante el uso de halogenaciones directas y secuenciasdecaptura de desprotolitiación como pasos clave.
